Cataract is a disease in which the crystalline lens becomes opacified causing a decline
in visual acuity. While many types of cataract are known, senile cataract is the most
Numerous theories have been advanced regarding the pathogenesis of senile cataract,
but neither preventive nor therapeutic modalities were available. Until recently, the
only course of action was to wait for the cataract to mature and then excise the opacified
lens. Under those circumstances, a compound having a pyrido-phenoxazine nucleus,
which structurally resembles xanthommatin, an insect eye pigment, was synthesized in
the laboratory of Senju. This compound has been found, by preclinical and clinical
studies, to prevent denaturation of the water-soluble protein of the lens and thereby
arrest the progression of cataract. Thus, for the first time, we have seen the advent of
pharmacotherapy for senile cataract.
